The study identifies the essentiality of the birA gene encoding biotin protein ligase (Mt BPL) in Mycobacterium tuberculosis and Mycobacterium smegmatis, and characterizes a bisubstrate inhibitor, Bio-AMS, that effectively targets Mt BPL, showing potent antitubercular activity against drug-resistant strains.
